Sermorelin

Pharmacological action. Synthetic peptide (protein), corresponding to the structure of the N-terminal fragment of the endogenous (produced in the body)-releasing factor in human growth hormone (hypothalamic neurohormones that promotes release of pituitary growth hormone). In action similar sermorelin-releasing factor, growth hormone, that is  interacting with specific membrane receptors, stimulates the synthesis and secretion (release) of growth hormone by the pituitary gland (endocrine gland, located in the brain).
After intravenous administration of 1.0 mg / kg concentration of growth hormone in blood serum increases to a maximum value after 10-60 min and decreases to its original level within 2-3 h.

Indications for use. Evaluation of functional capacity and responsiveness of anterior pituitary growth hormone (diaganostic method for suspected growth hormone deficiency in stunted children).

Dosage and administration. Doses are determined individually, depending on body weight of the studied individuals.

The drug is diluted immediately before use not less than 0.5 ml of the supplied solvent.

The drug at a dose of 1.0 mg / kg intravenously quickly (within 30 sec) in the morning on an empty stomach. Samples of venous blood taken 15 minutes before drug administration, immediately before its introduction and after 15, 30, 45, 60 and 90 min after injection.

If growth hormone levels in the blood after injection of 7 ng / ml or less, have stunted the growth of patients the cause of violations can be considered as growth hormone deficiency. If growth hormone levels in the blood after the injection of more than 7 ng / ml, the above cause dysplasia is unlikely to apply provocative tests to rule out hypothalamic dysfunction (dysfunction of the hypothalamus - endocrine glands).

Before the test  with sermorelinom cancel drugs, which have a direct effect on pituitary secretion of somatostatin: cyclo-oxygenase inhibitors (nonsteroidal anti-inflammatory drugs such as aspirin or indomethacin), means having in its composition or induce the release of somatostatin somatostatin, insulin (hormones of the adrenal cortex that affect carbohydrate and protein metabolism).

Concentration of growth hormone may increase under the influence of clonidine and levopy, as well as insulin-induced hypoglycemia (insulin-induced reduction in blood sugar levels). Cholinergic drugs in general enhance the effects of sermorelin and anticholinergic-tools reduce this effect.

On the test results may be affected: the simultaneous growth hormone therapy (in this case the test is carried out with sermorelin 1-2 weeks after discontinuation of therapy), the presence of untreated hypothyroidism (thyroid gland) or both antithyroid drugs therapy, a high concentration of somatostatin during the injection , obesity, hyperglycemia (high blood sugar) and high concentration of fatty acids in blood plasma? (Response to growth hormone administration sermorelin is lowered).

Side effects. Hot flashes and feeling a rush of blood  on the face, pain at the injection site.

Contraindications. Hypersensitivity to the drug, pregnancy, breast-feeding.

Release form. Dry substance for injection 50 mg in the ampoule in a package of 1 piece, complete with an ampoule of solvent (1 ml 0.9% sodium chloride).
